Abstract
|
|
|
Purpose: To identify and characterize of SH2 and SH3 domains has involved studying their roles in proteins produced by oncogene, The monoclonal antibodies against those domains was produced.
Meterials and Methods: To study aberrant SH2 interactions, SH2 domain of PLC-γ1 was purified using GST fusion system and immunized to mouse. The monoclonal antibodies was produced against SH2 domain by standard hybrodoma technology.
Results and Conclusion: The antibody recognized specifically to N-terminal SH2 domain of PLC-γ1. The monoclonal antibody, clone 4 precipitated SH223 domain as well as native PLC-γ1 in PC12 cell lysate overexpressed SH223 domain of PLC-γ1. Taken together, This monoclonal antibody can be used studying their roles in SH2 domain containing proteins produced by oncogens and proteins associated with aberrant SH2 interaction.
